Experimental Systems in Use


Dr Lee's lab uses the embryonal carcinoma cell line P19CL6 for preliminary studies of cardiac differentiation and proliferation. This cell line has the capacity to differentiate efficiently into cardiac myocytes following induction with the drug dimethyl sulfoxide or DMSO. As shown in the figures below, the expression of early markers of cardiac differentiation is activated as early as 4 days following induction (left); After 12 days of culture the cells are predominantly cardiac myocytes as demonstrated by staining for cardiac myofibrillar proteins (MF-20) and transcription factors (Gata4) (right). These cardiac myocytes appear fully functional, are electrically coupled to one another, and beat in synchronized fashion (see movies at high and low magnification).
